InvivoGen's screening assays enable the reliable detection of activators or inhibitors across key innate immune pathways, including Toll-like receptors (TLRs), NOD-like receptors (NLRs), C-type lectin receptors (CLRs), Stimulator of interferon genes (STING), Retinoic acid-inducible gene I (RIG-I), and Melanoma differentiation-associated protein 5 (MDA5). These sensitive assays are based on engineered HEK293, THP-1, or A549 cells expressing one or two inducible reporter systems: an NF-κB-inducible SEAP (secreted embryonic alkaline phosphatase) and/or an IRF (interferon regulatory factor)-inducible secreted Lucia® luciferase. InvivoGen's proprietary methods detect NF-κB and/or IRF pathway activation or inhibition by your compounds of choice.

Level 1 Compound Profiling TLR Response Profile. Engineered HEK-Blue™ reporter cells were stimulated with one fixed concentration of the client's compound "Compound X" or a positive control ligand*. After 24h incubation, TLR-induced NF-κB activation was assessed by measuring the levels of SEAP in the supernatants of HEK-Blue™ cells using QUANTI-Blue™.

Service options

InvivoGen offers two flexible screening levels, which can be ordered sequentially or individually, depending on your research needs. 

  • Level 1 - Compound Profiling:

Single-dose testing across one or more selected panels of PRRs to evaluate initial activity. Testing is performed in triplicate, typically at a 1/10 dilution or a concentration specified by the client.

  • Level 2 - Compound Dose Response:

Multi-dose testing on specifically chosen PRRs to assess dose dependency. Common concentrations include 1/10, 1/100, and 1/1000 dilutions, with further customization available upon request.

1. Reach out to tech@invivogen.com to discuss your project needs. Complete our Screening Request Form and our team will guide you in selecting the most relevant PRR panel for your therapeutic candidates.

2. We will provide a price quote for your selected screening services. Our lab will review the request and follow up with any questions or clarifications. 

3. Once your panel selection and quote are confirmed, send your samples to us. We’ll handle the preparation and set up your compounds for the screening.

4. Our PRR screening services use engineered cell lines in a controlled 96-well format, testing each compound in triplicate to ensure consistency and reliability.

5. After running the assays, our specialists prepare a finalized written report

6. Our team is available to discuss the results and offer insights on potential next steps in your drug discovery process.

Q: How much does the screening service cost?

The price depends on many factors, such as the screening level, number of compounds and cell lines, concentration ranges, and grade of customization. The indicated prices (see above) cover:

- Compound Profiling (Level 1): 1 compound at 1 concentration on our TLR panel (7 cell lines) + control cell line.

​- Compound Dose Response (Level 2): 1 compound at 5 concentrations on 1 PRR cell line + control cell line.

Q: How many concentrations should be tested to determine EC50 or IC50?

In our experience, a minimum of 7 concentrations be tested with a series of 1:3 dilutions to sufficiently calculate an EC50 or IC50.
